Police said officers had been called at about 20:00 BST to reports of the crash at Culver Down, near Bembridge.
Emergency crews said two bodies had been recovered at the bottom of the cliffs. Coastguard and fire and rescue teams joined police and ambulance officers at the scene.
Police have started an inquiry into the circumstances behind the crash.
They urged any witnesses to come forward.
Anne Ricketts, 47, from nearby Sandown, was walking her dog along the beach around the time of the incident.
She said she saw a tractor dragging a trailer containing four firefighters towards the beach.
Just after 20:00 she saw a coastguard helicopter in the area, while "truckloads" of rescuers from the coastguard were being dropped off nearby, she said.
